Immunopharmacology Comments
Lenalidomide is an immunomodulatory drug that binds the cereblon protein. It alters the substrate specificity of the CRL4CRBN E3 ubiquitin ligase, leading to selective ubiquitination and degradation of the lymphoid transcription factors Ikaros (IKZF1) and Aiolos (IKZF3), which ultimately elevates IL-2 production. In addition, casein kinase 1α (CK1α; CSNK1A1) is targeted for degradation by lenalidomide [2,4].
